Her first public appearance took place at the age of three in her father's church, where she recited the prayer "Our Father". Her family later moved to St. Louis, Missouri, where she spent her childhood. In St. Louis, Agnes Moorehead performed in the city's opera choir, where she was nicknamed "Mooney."

She graduated from high school in 1918, and although her father did not oppose his daughter's desire to become an actress, he still insisted that she graduate. Moorehead attended Muskingum College in New Concord, Ohio, graduating in 1923 with a bachelor's degree in biology. While studying in college, she first began performing on stage in the student theater troupe. Then Agnes Moorehead studied at the graduate school of the American Academy of Dramatic Arts, which she graduated in 1929.

Agnes Moorehead's early career was not very successful - she could not find a job for a long time, and only with great difficulty got a job as a radio host. She soon met actress Helen Hayes, who unsuccessfully tried to push her into the cinema, but in the end, Moorehead had to continue working on the radio. In 1937, Agnes Moorehead joined the Orson Welles theater company. Two years later, Wells signed a contract with RKO Studios and transferred his actors to Hollywood for filming. There Agnes Moorehead made her big screen debut in Wells' drama Citizen Kane (1941), where she played his mother.

A year later, the second film with the participation of the actress, "The Magnificence of the Ambersons," was released, for her role in which she was awarded a New York Critics' Award and was also nominated for an Oscar for Best Supporting Actress. This was followed by roles in such films as "Journey into Fear" (1943), "Miss Parkington" (1944), which brought her a second Oscar nomination, "Since You Left" (1944), "Johnny Belinda (1948), her third Academy Award nomination, and Floating Theater (1951).

During the 1940s and 1950s, Agnes Moorehead was one of the most sought-after actresses on radio, theater and cinema. In 1951-1952 she had a role in the Broadway production of Don Juan in Hell, and from 1962 to 1963 she played in the play Lord Pengo.

In 1964, the actress became the performer of the role of the sorceress Endora, the mother of the main character, in the television series "My Wife Bewitched Me". This role brought Agnes Moorehead great popularity, and she did not part with her until the end of the show in 1972. Throughout her television career, she has received Emmy nominations six times, but only once won her for her role in the drama series Wild Wild West. The fourth and last time the actress was nominated for "Oscar" in 1964 for the role of Velma Crater in the film "Hush, Hush Sweet Charlotte." She did not receive the Academy Award, but she won another award - the Golden Globe.
