Shankman was born in New York to Shepard A. Shankman, who worked for a consulting company, and his wife Katherine, a lawyer at a law firm. He graduated from Brown University and then received his Master of Fine Arts from New York University in 1993, completing the acting program at the Tisch School of the Arts. Shankman's sister Elizabeth is married to British publicist Jamie Byng, with whom she has two children: Ivy and Nathani.

Since 2005, Shankman has been married to Lauren Bennett Greilshemer.

Shankman began his professional acting career with a small role in Robert Redford's 1994 film Trivia and a guest role on the television series Law & Order, which was the first of seven appearances for the actor on the show. He also began working at the American Conservatory Theater in San Francisco, playing the role of Luis Irons in Tony Kushner's Angels in America.

While at New York University, Shankman played the role of Roy Cohn in a studio production of this play, and eight years later he again played the role of Louis Irons in the HBO miniseries Angels in America, receiving Emmy and Golden Globe nominations.
