In 2005, Jemima made her debut as a voice actress. She voiced the twin singer in Smile for the Camera .
In 2010, Jemima played the role of Charlotte in her director friend Lina Dunham's Tiny Furniture, for which she received a nomination for the 2010 Gotham Independent Film Awards. Kerk starred in Tiny Furniture for free because there wasn't enough money to make it. Despite the fact that the film proved to be profitable, Jemima was not paid.
In 2011, Kirk starred in the music video for Rival Schools, her friend's group, for the song Wring It Out.
Since 2012 Jemima has been playing the role of Jessa Johansson in the television series Girls, also directed by Lina Dunham.
